I didn't take it as "weirdly defensive" in the least. I took Nicole's emphatic "no" as keeping things from getting any messier, with Brad hitting on a lesbian who is also Ruby's ex and is also crazy. And I definitely didn't see that interaction as showing us that Nicole thinks Cheyenne is hot and it annoys her. (I think Nicole's entire interaction with Cheyenne was probably showing us that Nicole thinks she's hot and it annoys her, though.)

You know, this kind of confirms that Ruby has a "type". Pretty, feminine, long hair, big personality, and a little bit crazy... Sure, Nicole and Cheyenne aren't crazy in the same way--Nicole isn't a psychopath--but she's got the big ego, the snippy, sassy, traits and she can be a bit selfish and manipulative in small ways. (And I love her to bits!)
